cft ceramic fluoroapatite (ca10(po4)6(f2) is a composite of fluoroapatite and hydroxyapatite prepared by converting hydroxyapatite nanocrystals with a fluorine reagent.
cft has separation characteristics similar to those of cht ceramic hydroxyapatite, and can be used under stringent chromatography conditions to separate acidic proteins requiring buffered conditions as low as ph 5, with minimal compromise of the solubility or lifetime of the support.
cft is a rigid macroporous support and is available in two distinct material types, type i and type ii, and in one particle size, 40 µm. it has high binding capacity and may be used reproducibly over an extended number of cycles. its increased tensile strength, chemical durability, and density enable it to provide the throughput and consistent performance for all separations, including biopharmaceutical process-scale manufacturing.
cft type i and type ii are chemically identical, but have been sintered at different times and temperatures, resulting in physically and chemically stable media. the dynamic capacity is higher for cft type ii than type i, the opposite of cht.
protein se-lectivity is expected to be unaffected regardless of use of cft or cht. solubility or apatite lifetime, however, will be less of a concern with cft when lower ph buffered systems are a requirement. in order to determine which material types provide optimal chromatographic performance, evaluation of all four options — cft and cht, both types i and types ii — should be performed side by side.
